I am installing a toekick heater in a bath remodel. This replaces an old cast iron hot water unit. My question is can I connect the booster circulator to the return line instead of the supply line? I have space constraints on the supply line.

I would make sure
that the toe-kick heater has it's own zone and priority. When coupled to cast iron and the cast iron governs the building control, you will be blowing cold air as often as not. (The cast iron "coasts" between firings; the toe-kick heater is like fin-tube, heating and cooling almost instantly with no mass.)
Putting such a small load on a system and giving it priority is a case of the tail wagging the dog, however.0
