How can I pipe a 40 plate heat exchanger to radiant floor? Heat have to connecting rooms both on their own manifolds, same thermostat. Rooms are open to each other one is 280sqft, other is 800sqft. Can i combine manifolds and use one pump and 3 way valve or do I have to run them separate to heat exchange on their own pump and 3 way valve?

Assuming you have the HX sized to the flow required and the pump sized to the loads on the B side, here is a simple example.
Watch the Cv on thermostatic mix valves, they are fairly restrictive valves, probably 5-6 gpm max on the typical 1", 3 Cv valves.
Of course you need an air eliminator, expansion tank, relief valve, purge ports, iso valves, etc on both sides of the HX.Bob "hot rod" Rohr
